Neutrophils play an important role in cancer disease progression and due to their plasticity, they can have different functions in the tumor micro-environment, with both pro and anti-tumor properties. Neutrophils have been associated with multiple functions, such as killing antibody-opsonized cancer cells, directly killing of tumor cells through trogocytosis, the release of reactive oxygen and nitrogen species. Studying of neutrophil functions, such as antibody-dependent cell-mediated cytotoxicity (ADCC), Neutrophil Extracellular Traps (NETs) and the release of ROS, and the screening of the potency of therapeutic candidate to activate neutrophils in vitro can be done using the Incucyte® live imaging system. Further, multiplex cytokines and peroxidase enzymes analysis after activation and stimulation of neutrophils can be evaluated using ELISA or Luminex technology. Finally, when studying neutrophil activity in vitro, important factors such as access to fresh blood and optimized protocols for purifying untouched neutrophils are key for reproducible and reliable results.
