Track 2: From Bench to Bedside: Bioanalysis and Translational Science
Advanced Multiplexing and Quantification Techniques for Biomarkers and Proteins Using HRMS/NanoLC
presented by Prakash Bhosale, Ph.D. and Hemanth Akkiraju, Ph.D.
This presentation will cover the development and optimization of advanced multiplexing assays and quantification techniques for biomarkers and proteins using high-resolution mass spectrometry (HRMS). We will explore the simultaneous detection of ACTH and ANP, demonstrating ultra-sensitive readout capabilities and precise quantification at low concentrations. Label free analysis with nano-liquid chromatography (NanoLC) coupled to High-throughput readouts will be emphasized, showcasing efficient and reproducible methods for multiplexing of peptide hormone biomarkers analysis. We will address the challenges of quantifying proteins exhibiting multiple charge states, providing practical solutions for accurate and reproducible quantification. Additionally, the use of Tandem Mass Tag (TMT) labeling for enhanced quantification of proteins will be included, highlighting its benefits for multiplexed quantification and increased throughput. Attendees will gain a comprehensive understanding of these advanced techniques and their applications in clinical diagnostics and research, along with insights into best practices and potential challenges.
Learning objectives:
- Label free peptides analysis
- Advantages of nano-LCMS
- TMT labeling for quantification of proteins
